报告摘要:Covalent chemistry can be observed at the single-molecule level by using engineered protein pores as “nanoreactors”. By recording the ionic current driven through single α-hemolysin nanoreactors under a transmembrane potential, individual bond-making and bond-breaking steps are monitored with sub-millisecond time-resolution. Covalent chemistry observed by this approach has included multiple turnovers of reversible reactions, polymer chain elongation, and complex reaction networks. The scope of the approach can be expanded by using unnatural amino acid mutagenesis to place a variety of reactive side chains within the lumen of the pore. Recently, we have turned our focus from observing chemistry at the single-molecule level to manipulating chemistry, especially that of polymers, by exploiting the environment within a nanopore, notably confinement. Site-selective chemistry has been effected on polymer substrates under confinement by spatial alignment of the reactive groups. We also built a highly processive mobile molecule, termed a “hopper”, which chemically takes sub-nanometre steps within a confined space while carrying a polymer cargo. This chemistry is being applied to the stepwise translocation of biopolymers for sequencing applications and for selective transmembrane signalling in synthetic tissues.
